ABSTRACT

The objective of this work was to evaluate the biochemical and hormonal variables of Moxotó and Canindé goats submitted to two temperatures - 26.0±0.6 (thermoneutral) and 32.0±1.2°C (above thermal comfort zone) - and consuming water with three levels of salinity (1.0, 6.0 and 12.0 dSm-1). Thirty-six animals (18 of each breed) were used, with an average age of 5.0±0.6months and an average weight of 20.0±2.3kg, housed in metabolic cages inside a climate chamber. The animals were distributed in a completely randomized design with a 2 × 2 × 3 factorial scheme (2 breeds, 2 temperatures and 3 levels of salinity) and three replications. The glucose and urea had a significant effect (P>0.05) according to water salinity. Glucose, cholesterol, protein, albumin, globulin, aspartate aminotransferase and hormones (T4, T3 and cortisol) varied according to temperature (P<0.05). There was a significant effect of time on hormonal variables (P<0.05). Biochemical and hormonal variables changed according to temperature and day shift, so that metabolism was reduced in the animals under thermal stress and accelerated when animals were in the thermal comfort zone.(AU)


O objetivo do trabalho foi avaliar as variáveis bioquímicas e hormonais de caprinos das raças Moxotó e Canindé, submetidos a duas temperaturas (26,0±0,6ºC e 32,0±1,2ºC), termoneutra e acima da zona de conforto térmico, respectivamente), consumindo água com três níveis de salinidade (1,0, 6,0 e 12,0dSm-1), utilizando-se 36 animais (18 de cada raça), com idade média de 5,0±0,6 meses e peso médio de 20,0±2,3kg, alojados em gaiolas metabólicas no interior de uma câmara climática. Os animais foram distribuídos em um delineamento inteiramente ao acaso, com esquema fatorial de 2 x 2 x 3 (2 raças, 2 temperaturas e 3 níveis de salinidade) e três repetições. A glicose e a ureia apresentaram efeito significativo (P<0,05) em função da salinidade da água. Glicose, colesterol, proteína, albumina, globulina, AST e hormônios (T4, T3 e cortisol) variaram conforme as temperaturas (P<0,05). Observou-se efeito significativo do horário sobre as variáveis hormonais (P<0,05). As variáveis bioquímicas e hormonais sofrem alterações em função da temperatura e do turno do dia, de modo que o metabolismo é reduzido em animais sob estresse térmico e acelerado quando os animais encontram-se na zona de conforto térmico.(AU)

ABSTRACT

The crab Ucides cordatus is an important feature of mangroves in the North and Northeast of Brazil. In 2002 a large mortality was reported on this species in the Jaguaribe River Estuary. In order to investigate the possible causes specimens were collected monthly between May 2002 and December 2003 at five sites: Fortim (E1), Fortim (E2), Cumbe (E3) Aracati (E4) and Guajiru (E5). The mangrove structure and environmental parameters (salinity, pH, temperature and DO) were also evaluated. Water was collected for suspended solids analysis, BOD and nutrient levels. Salinity decreased in the rainy season, with no significant variation of pH and temperature. Dissolved oxygen levels had a mean of 5.7 mg L-1. Suspended solids increased at site E4, with BOD varying at E1 and E2 during the dry season. The same was observed at sites E1, E2 and E3 during the rainy season. Ammonia levels at E1 and E2 and phosphorus in all seasons, increased with rain events. Four lines of investigation were adopted: biometry, histology, hematology, and bioassay. There was no significant difference between animals in biometrics and histology showed no cellular alterations. However, hematology showed a significant difference between the E1 and E3 sites due to depletion in the number of hemocytes in E1, probably in response to environmental impacts. This can lead to poor immunity, leading to opportunistic pathogen infections such as viruses, bacteria and fungi. The bioassay showing no abnormal behavior or mortality. The structure the of mangrove was developed in all areas, except in E5 which served as a control site, with significant environmental stress with high levels of herbivorous growth (> 50%) and a salinity of around 50 ‰. It is believed that the mortality of the crabs was an occasional occurrence, probably due to toxin production by some fungal organism.

A mitilicultura no Brasil se baseia no mexilhão Perna perna (Linnaeus, 1758), que é encontrado em todo o litoral brasileiro, sendo especialmente abundante do Espírito Santo a Santa Catarina. Nos últimos anos, foi lançada a hipótese de que P. perna seja uma espécie exótica no litoral brasileiro. A hipótese se baseia na análise da malacofauna presente em sítios arqueológicos. Todas as contestações levantadas não remetem à uma conclusão, pois precisam de estudos específicos, com metodologias claras, aliando arqueologia, ecologia e biologia molecular. O objetivo deste trabalho foi estudar a condição de nativo ou exótico do mexilhão Perna perna no Brasil, a partir de levantamento dos resultados em sítios arqueológicos, de técnicas moleculares e de datação de conchas com C14. A datação indicou que as amostras de Perna perna do sítio arqueológico Rio do Meio/Jurerê, Florianópolis/SC, têm idade de 720±30 e 780±30 anos. O cálculo do tempo de divergência indicou que a separação das populações brasileiras e africanas ocorreu por volta de 200 mil anos. Os resultados apontam a presença da espécie no território brasileiro muito antes do descobrimento do Brasil pelos portugueses no ano de 1500, indicando que P. perna é de fato uma espécie nativa.


Mussel farming in Brazil is based on brown mussel Perna perna (Linnaeus, 1758), which is found throughout the Brazilian coast, especially abundant from Espírito Santo to Santa Catarina coast. In recent years it was suggested that Perna perna is an exotic species for the Brazilian coast. The hypothesis is based on the analysis of the zooarchaeology studies in archaeological sites in Brazil. All objections raised do not offer a conclusion, because they need specific studies with clear methodology, combining archeology, ecology and molecular biology. The aim of this work was to study the condition of the brown mussel Perna perna in Brazil if native or exotic, from survey results in archaeological sites, molecular techniques and dating of shells with C14. The dating indicated that the shells were 720±30 and 780±30 years, respectively. The calculation of divergence time indicated that the separation of the African and Brazilian mussel populations occurred around 200 thousand years ago. The results indicate the presence of the P. perna species in Brazilian territory long before the discovery of Brazil by the Portuguese in 1500, indicating that P. perna is actually a native species of Brazil.

ABSTRACT

Treatment and safe disposal of tannery saline wastewater, a primary effluent stream that is generated by soaking salt-laden hides and skin is one of the major problems faced by the leather manufacturing industries. Conventional treatment methods like solar evaporation ponds and land composting are not eco-friendly as they deteriorate the ground water quality. Though, this waste stream is comprised of high concentration of dissolved proteins the presence of high salinity (1-6 percent NaCl by wt) makes it non-biodegradable. Enzymatic treatment is one of the positive alternatives for management of such kind of waste streams. A novel salt-tolerant alkaline protease obtained from P.aeruginosa (isolated from tannery saline wastewater) was used for enzymatic degradation studies. The effect of various physical factors including pH, temperature, incubation time, protein source and salinity on the activity of identified protease were investigated. Kinetic parameters (Km , Vmax) were calculated for the identified alkaline protease at varying substrate concentrations. Tannery saline wastewater treated with identified salt tolerant protease showed 75 percent protein removal at 6 h duration and 2 percent (v/v) protease addition was found to be the optimum dosage value.

ABSTRACT

Aim: To investigate the casuistry of drowning cases by reviewing the records from the Forensic Medicine Institute Nina Rodrigues in the city of Salvador, BA, Brazil, and to verify the potential of DNA recovery in human teeth immersed in water. Methods: An epidemiological survey was conducted followed by a laboratorial phase, in which 40 teeth were immersed in fresh and salt-water, the DNA was extracted by the organic method and amplified by polymerase chain reaction, using the amelogenin as initiator. The electrophoresis initially occurred in agarose gel and later in polyacrylamide gel. Results: In the present survey, 346 deaths from drowning were observed, most of them in salt-water (51.73%), with a predominance of male victims (86.13%) aged from 18 to 35 years-old (37.94%). Dentists identified 14.74% of the victims. DNA was recovered in 37.5% from the samples, most from teeth immersed in freshwater. Polyacrylamide gel analysis in samples that were amplified in agarose gel allowed correct gender identification in 83.3% of the cases. However, allele loss was observed in samples of two victims, jeopardizing gender determination. Conclusions: Dental exposure to water interfered in DNA recovery. The gender investigation using the amelogenin as initiator was effective.
